Italy’s Matteo Arnaldi was infected with the virus minutes before the semi-finals, while Flavio Cobolli advanced to the finals of the French Open.
The best friends were due to meet in the last four on Friday at 18:00 BST to take part in their first Grand Slam final.
However, Arnoldi’s suspension was announced 20 minutes before the players kicked Philippe Chatrier on the court.
World No. 14 Kobolli has put him straight into Sunday’s Roland Garros showdown, where he will face second seed and favorite Alexander Zverev.
Arnoldi spent 19 hours and 42 minutes on court en route to the semi-finals in Paris – the most since 1991 when anyone advanced to the last four of a major.
